The Agilent’s analyzer, Seahorse Xfe24, measures the oxygen consumption rate (OCR) wich is an indicator of mitochondrial respiration and the extracellular acidification rate (ECAR) in the time (intervals of approximately 5 to 8 minutes).

OCR is an, and ECAR mainly is the result of glycolysis.

Real-time measurements of OCR and ECAR are performed by isolating a very small volume (approximately 2 µL), also called a "transient microchamber," of medium on top of a monolayer of cells in a microplate.

The consumption of cellular oxygen (respiration) and the release of protons (glycolysis) cause rapid and easily measurable changes in the concentrations of dissolved oxygen and free protons in this "transient micro-chamber" which are measured every few seconds by probes located 200 microns above the cells. The instrument measures changes in concentrations for 2 to 5 minutes, then calculates OCR and ECAR, respectively.

When the measurement is complete, the probes lift up, allowing the large volume of medium to mix in the transient microchamber, thereby restoring cell values ​​to their baseline level. An integrated drug delivery system allows up to four compounds to be sequentially added per well at user-defined intervals.

This technology allows the study of bioenergetic changes in real time allowing a broad functional study.


 

 

Since Seahorse's measurements are non-destructive, the metabolic rate of the same cell population can be repeatedly measured over time, while four different drugs are injected sequentially into each well.

The total duration of the test is usually 60 to 90 minutes.
